Japonya'nın en prestijli ulusal üniversitelerinden biri olan Osaka Üniversitesi 1931 yılında kurulmuştur ancak kökeni Japonya'da Batı tıbbının öncülerinden Ogata Kōan tarafından 1838 yılında kurulan Tekijuku Okulu'na dayanmaktadır.

Japonya'nın imparatorluk döneminde 1931 yılında resmi olarak Osaka İmparatorluk Üniversitesi adını almış, İkinci Dünya Savaşı'nın ardından 1947 yılında bugünkü adını almıştır.
